چکیده

Murine double minute 2 (MDM2) and 4 (MDM4) are known as the main negative regulators of p53, a tumor suppressor. They are able to form heterodimers that are much more effective in the downregulation of p53. Therefore, the MDM2-MDM4 complex could be a target for promising therapeutic restoration of p53 function. To this aim, a deeper understanding of the molecular mechanisms underlining the heterodimerization is needed. The kinetic and thermodynamic characterization of the MDM2-MDM4 complex was performed with two complementary approaches: atomic force spectroscopy and surface plasmon resonance. Both techniques revealed an equilibrium dissociation constant (KD ) in the micromolar range for the MDM2-MDM4 heterodimer, similar to related complexes involved in the p53 network. Furthermore, the MDM2-MDM4 complex is characterized by a relatively high free energy, through a single energy barrier, and by a lifetime in the order of tens of seconds. New insights into the MDM2-MDM4 interaction could be highly important for developing innovative anticancer drugs focused on p53 reactivation.

Behenic Acid Monolayer and Bilayer Assemblies- A Study of Concanavalin A (Con A) Adsorption and its Interaction with Dextran Using Surface Plasmon Resonance Spectroscopy and Microscopy

Deposition of behenic acid (B.A) mono and bilayers onto gold coated surfaces was performed by Langmuir-Blodget dip-casting technique. Surface Plasmon resonance (SPR) and ellipsometry methods were employed for investigation of the monolayer and bilayer films. The adsorption of the biologically important molecule Concanavalin A (Con A) from bulk solution to these monolayers and bilayers as we...

Imaging and kinetics of the bimolecular complex formed by the tumor suppressor p53 with ubiquitin ligase COP1 as studied by atomic force microscopy and surface plasmon resonance

p53 plays an important role in the safeguard of the genome but it is frequently downregulated mainly by E3 ubiquitin ligases among which COP1 plays an important role. The overexpression of COP1 has been reported to occur in several tumors and may be indicative of its overall oncogenic effect, which in turn might be originated by a direct interaction of COP1 with p53. Such an interaction may con...

Formation and Characterization of Self-Assembled Phenylboronic Acid Derivative Monolayers toward Developing Monosaccharide Sensing-Interface

We designed and synthesized phenylboronic acid as a molecular recognition model system for saccharide detection. The phenylboronic acid derivatives that have boronic acid moiety are well known to interact with saccharides in aqueous solution; thus, they can be applied to a functional interface of saccharide sensing through the formation of self-assembled monolayer (SAM). In this study, self-ass...

Molecular Recognition of Arginine by Supramolecular Complexation with Calixarene Crown Ether Based on Surface Plasmon Resonance

Arginine plays an important role in cell division and the functioning of the immune system. We describe a novel method by which arginine can be identified using an artificial monolayer based on surface plasmon resonance (SPR). The affinity of arginine binding its recognition molecular was compared to that of lysine. In fabrication of an arginine sensing interface, a calix[4]crown ether monolaye...

Fabrication and Characterization of the Fiber Optical Taper for a Surface Plasmon Resonance Sensor

For a fiber optical surface plasmon resonance (SPR) sensor a short part of its cladding should be removed to coat a thin layer of a metal. Usually this is problematic when an optical fiber with small core diameter is used. In this paper, a new method using µliter droplet of the HF acid for short fiber optical taper fabrication is reported. Using this method in a multi-mode optical fiber w...
